Abstract

ABSTRAK Xylaria sp. merupakan fungi penghasil enzim kitinase yang berperan dalam degradasi kitin dan berpotensi dimanfaatkan sebagai agen pengendalian hayati. Aktivitas enzim dipengaruhi oleh berbagai faktor lingkungan, salah satunya suhu. Penelitian ini bertujuan untuk menganalisis pengaruh variasi suhu terhadap aktivitas enzim kitinase dari Xylaria sp. serta menentukan suhu optimumnya.a Penelitian ini menggunakan pendekatan kuantitatif dengan desain eksperimen laboratorium dengan Rancangan Acak Lengkap (RAL) yang terdiri atas enam perlakuan suhu, yaitu 25°C, 27°C (kontrol), 30°C, 35°C, 40°C, dan 45°C dengan tiga kali ulangan. Aktivitas kitinase dianalisis secara kualitatif menggunakan media CCA melalui pengamatan zona bening dan perhitungan indeks kitinolitik. Pengujian kuantitatif dilakukan menggunakan metode DNS (3,5-dinitrosalicylic acid) berdasarkan pengukuran gula pereduksi. Hasil penelitian menunjukkan bahwa variasi suhu berpengaruh terhadap aktivitas enzim kitinase Xylaria sp. baik secara kualitatif maupun kuantitatif. Aktivitas kitinolitik tertinggi berdasarkan indeks kitinolitik diperoleh pada suhu 35°C dengan nilai 1,08, sedangkan aktivitas enzim kitinase tertinggi secara kuantitatif diperoleh pada suhu yang sama dengan nilai 0,239 ± 0,028 U/mL. Aktivitas enzim terendah diperoleh pada suhu 45°C sebesar 0,185 ± 0,003 U/mL. Berdasarkan hasil penelitian dapat disimpulkan bahwa suhu optimum aktivitas enzim kitinase Xylaria sp. adalah 35°C. Temuan ini menunjukkan potensi Xylaria sp. sebagai sumber enzim kitinase yang dapat dikembangkan untuk aplikasi bioteknologi dan pengendalian hayati yang ramah lingkungan. Xylaria sp. is a fungus that produces the enzyme chitinase, which plays a role in chitin degradation and has the potential to be used as a biological control agent. Enzyme activity is influenced by various environmental factors, one of which is temperature. This study aims to analyze the effect of temperature variations on the chitinase activity of Xylaria sp. and to determine its optimum temperature.a This study employed a quantitative approach using a laboratory experimental design with a Completely Randomized Design (CRD) consisting of six temperature treatments: 25°C, 27°C (control), 30°C, 35°C, 40°C, and 45°C, with three replicates. Chitinase activity was analyzed qualitatively using CCA medium through observation of clear zones and calculation of the chitinolytic index. Quantitative testing was performed using the DNS (3,5 dinitrosalicylic acid) method based on the measurement of reducing sugars. The results showed that temperature variations affected the chitinase activity of Xylaria sp. both qualitatively and quantitatively. The highest chitinolytic activity, as determined by the chitinolytic index, was obtained at 35°C with a value of 1.08, while the highest quantitative chitinase activity was obtained at the same temperature with a value of 0.239 ± 0.028 U/mL. The lowest enzyme activity was observed at 45°C, at 0.185 ± 0.003 U/mL. Based on the research results, it can be concluded that the optimum temperature for Xylaria sp. chitinase activity is 35°C. These findings indicate the potential of Xylaria sp. as a source of chitinase that can be developed for biotechnology applications and environmentally friendly biological control.
