Abstract
ABSTRAK Salah satu penyebab masalah utama di Tempat Pemrosesan Akhir (TPA) Provinsi Lampung adalah meningkatnya tumpukan sampah plastik yang dihasilkan oleh aktivitas manusia. Hal ini dapat menjadi sumber polusi mikroplastik dan membahayakan jika mengendap di dalam tubuh organisme. Jamur dapat menjadi alternatif dalam membantu proses biodegradasi alami dalam mengurangi pencemaran mikroplastik pada ekosistem tanah. Penelitian ini bertujuan untuk mengidentifikasi dan mengkarakterisasi isolat jamur yang memiliki kemampuan dalam mendegradasi mikroplastik di Tempat Pemrosesan Akhir (TPA) Provinsi Lampung. Metode pengambilan sampel dilakukan dengan purposive sampling. Koloni yang tumbuh diidentifikasi dengan pengamatan pada zona bening, kemudian dikarakterisasi secara morfologi. Analisis data dengan metode deskriptif kualitatif dan pengukuran indeks zona bening. Hasil dari penelitian ini berhasil mengidentifikasi 13 isolat jamur yang menunjukkan area zona bening. Empat isolat (Ba1.2, Ba2.5, Bu3.4, Ka2.3) mampu mendegradasi ketiga jenis mikroplastik. Zona bening tertinggi: Bu3.4 untuk PET (2,00 mm), Ka2.4 untuk PE (1,50 mm), serta Ba2.3 dan Ba2.5 untuk PP (1,25 mm). Identifikasi dan karakterisasi morfologi menunjukkan 8 isolat termasuk genus Aspergillus, 3 isolat Penicillium, 1 isolat 1 Fusarium, dan 1 isolat Sclerotium. Degradasi terjadi melalui adhesi hifa dan sekresi enzim pemecah polimer. Kata kunci: biodegradasi, indeks zona bening, jamur, mikroplastik, Tempat Pemrosesan Akhir. ABSTRACT One of the main causes of problems at the Final Disposal Site (TPA) in Lampung Province is the increasing accumulation of plastic waste generated by human activities. This can become a source of microplastic pollution and be harmful if it accumulates in the bodies of organisms. Mushrooms can be an alternative in aiding the natural biodegradation process to reduce microplastic pollution in soil ecosystems. This study aims to identify and characterize fungal isolates that have the ability to degrade microplastics at the Final Disposal Site (TPA) in Lampung Province. The sampling method was conducted using purposive sampling. The colonies that grow are identified by observing the clear zones, then characterized morphologically. Data analysis using qualitative descriptive methods and measurement of the clear zone index. The results of this study successfully identified 13 fungal isolates that showed clear zones. Four isolates (Ba1.2, Ba2.5, Bu3.4, Ka2.3) are capable of degrading all three types of microplastics. The highest clear zone: Bu3.4 for PET (2,00 mm), Ka2.4 for PE (1,50 mm), and Ba2.3 and Ba2.5 for PP (1,25 mm). Identification and morphological characterization showed 8 isolates including the genus Aspergillus, 3 isolates of Penicillium, 1 isolate of Fusarium, and 1 isolate of Sclerotium. Degradation occurs through hyphal adhesion and the secretion of polymer degrading enzymes. Keyword: biodegradation, clear zone index, fungi, landfill, microplastics.
