Abstract

ABSTRAK Bakteri selulolitik halofilik mampu mendegradasi selulosa di lingkungan dengan salinitas tinggi (2%-30% atau >4 dS/m). Bakteri selulolitik halofilik mampu bertahan pada lingkungan salin dengan menghasilkan enzim selulase. Enzim selulase dapat berperan penting dalam berbagai industri seperti pangan, detergen, tekstil, bahan bakar, dan pengolahan limbah. Penelitian ini bertujuan mengeksplorasi nilai daya hantar listrik (DHL), pH tanah, serta kelimpahan dan karakteristik bakteri selulolitik halofilik di tanah. Penelitian ini bersifat eksploratif secara kuantitatif-deskriptif dengan menggunakan purposive sampling dalam menentukan pengambilan sampel. Pengumpulan data diawali dengan tahap persiapan, survei, pengambilan sampel tanah, dan pengujian di laboratorium. Analisis data menggunakan uji analysis of variance (ANOVA) batas kepercayaan yaitu 95% (α = 0,05) yang dilanjutkan dengan uji Duncan. Hasil penelitian menunjukkan tanah sentra industri ikan asin tergolong salin-sodik, dengan salinitas tertinggi berasal dari sampel 1 yang memiliki nilai DHL 1736 dS/m dan pH tertinggi berasal dari sampel 2 dengan pH sebesar 8,36. Kelimpahan bakteri tertinggi berasal dari sampel 1 yaitu 7,1x106 CFU/ml yang tumbuh pada media CMC 0,5% dengan penambahan NaCl 2%. Berdasarkan hasil isolasi dan karakterisasi secara makroskopis diperoleh 19 jenis isolat yang berbeda, dimana terdapat 5 isolat atau 26,32% yang mampu mendegradasi selulosa dengan sel berbentuk basil dan berwarna ungu (gram positif). Indeks aktivitas enzimatik tertinggi dihasilkan dari isolat B17.2 yaitu sebesar 1,12 cm, dan nilai aktivitas spesifik enzimatik tertinggi dihasilkan oleh isolat B17.11 yaitu sebesar 127,46 U/mg. Kelima Isolat bakteri selulolitik halofilik memiliki pertumbuhan optimal pada cekaman salinitas 4%, sehingga termasuk dalam kategori halofilik ringan. Pertumbuhan isolat bakteri ini berada dalam kondisi optimal pada pH 7 dan suhu 30ºC. Kata kunci: Bakteri seluloltik halofilik, enzim selulase, kelimpahan bakteri, salinitas, Pulau Pasaran. ABSTRACT Halophilic cellulolytic bacteria are capable of degrading cellulose in environments with high salinity (2%-30% or >4 dS/m). These bacteria survive in saline environments by producing cellulase enzymes. The cellulase enzyme plays a significant role in various industries such as food, detergent, textiles, biofuels, and waste management. This study aims to explore electrical conductivity (EC) values, soil pH, as well as the abundance and characteristics of halophilic cellulolytic bacteria in the soil. This research is exploratory and employs a quantitative descriptive approach using purposive sampling for sample collection. Data collection begins with preparation, surveys, soil sampling, and laboratory testing. Data analysis involves analysis of variance (ANOVA) at a 95% confidence level (α = 0.05), followed by Duncan’s test. The results show that the soil in the salted fish industry center is classified as saline-sodic, with the highest salinity from sample 1, which has an EC value of 1736 dS/m, and the highest pH from sample 2, with a value of 8.36. The highest bacterial abundance is found in sample 1, with 7.1×10⁶ CFU/ml, grown on 0.5% CMC medium with the addition of 2% NaCl. Macroscopic isolation and characterization identified 19 distinct isolates, 5 of which (26.32%) are capable of degrading cellulose. These isolates exhibit rod shaped cells and purple coloration (Gram-positive). The highest enzymatic activity index is produced by isolate B17.2 at 1.12 cm, while the highest specific enzymatic activity is achieved by isolate B17.11 at 127.46 U/mg. The five halophilic cellulolytic bacterial isolates exhibit optimal growth under 4% salinity stress, categorizing them as slightly halophilic.
