Abstract

ABSTRAK Bakteri selulolitik termofilik merupakan bakteri yang berhabitat pada suhu lingkungan antara 45°-65°C dengan pH 6-7. Bakteri ini mampu mengahasilkan enzim selulosa yang kini telah diproduksi secara komersial oleh berbagai industri global dan banyak dimanfaatkan dalam berbagai sektor seperti makanan, bahan bakar, industri kertas dan tekstil. Penelitian ini bertujuan mengkarakteristikkan dan mengukur indeks selulase bakteri selulolitik termofilik dari sumber air panas Way Panas Bumi Natar Lampung Selatan. Metode penelitian ini menggunakan metode kualitatif. Sampel diambil dari tiga titik wisata sumber air panas, adapun teknik yang digunakan dalam pengambilan sampel yaitu Purposive sampling dengan memperhatikan parameter fisika dan kimia. Hasil penelitian memperoleh 18 isolat yang berhasil diisolasi dari wisata sumber air panas, setelah di lakukan pewarnaan gram, 13 diantaranya merupakan gram negatif. Bentuk dominan isolat bakteri yang berhasil diisolasi yaitu berbentuk basil. Hasil uji aktivitas selulase didapatkan 5 isolat yang mampu mendegradasi selulase. Indeks selulase tertinggi yang didapat dari wisata sumber air panas yaitu 3,80 cm. Kata kunci: Bakteri termofilik, Carboxyl Methyl Cellulose (CMC), Selulolitik, Sumber air panas. ABSTRACT Thermophilic cellulolytic bacteria are bacteria that inhabit environments with temperatures ranging from 45°C to 65°C and a pH of 6-7. These bacteria can produce cellulase enzymes, which are now commercially produced by various global industries and widely utilized in sectors such as food, fuel, paper, and textiles. This study aims to characterize and measure the cellulase index of thermophilic cellulolytic bacteria from the Way Panas Bumi hot spring in Natar, South Lampung. The research employed qualitative methods. Samples were collected from three points in the hot spring tourist area using a purposive sampling technique, considering physical and chemical parameters. The study successfully isolated 18 bacterial isolates from the hot spring area. After Gram staining, 13 isolates were identified as Gram-negative. The dominant shape of the isolated bacteria was rod-shaped. Cellulase activity tests revealed that 5 isolates were capable of degrading cellulase. The highest cellulase index obtained from the hot spring was 3.80 cm. Keywords: Carboxyl Methyl Cellulose (CMC), Cellulolytic, Hot springs, Thermophilic bacteria.
